Histonmodifikációk refers to the biochemical modifications that occur on histone proteins. Histones are the main protein components of chromatin, the complex of DNA and proteins that forms chromosomes within the nucleus of eukaryotic cells. These modifications play a crucial role in regulating gene expression by altering the accessibility of DNA to the transcriptional machinery.

The most common histone modifications include acetylation, methylation, phosphorylation, ubiquitylation, and sumoylation. Each type of modification can occur on specific amino acid residues within the histone tails, which are the unstructured N-terminal extensions of histone proteins. For example, histone acetylation typically involves the addition of an acetyl group to lysine residues, which neutralizes their positive charge. This generally leads to a loosening of the chromatin structure, promoting gene transcription.

Histone methylation, on the other hand, involves the addition of methyl groups to lysine or arginine residues. Methylation can either activate or repress gene expression depending on the specific residue and the number of methyl groups added. Histone phosphorylation, the addition of a phosphate group to serine, threonine, or tyrosine residues, is also involved in chromatin condensation and gene regulation. Ubiquitylation and sumoylation are larger protein modifications that can affect histone stability and chromatin structure.

These modifications are not static; they are dynamically added and removed by specific enzymes, creating a "histone code" that dictates the state of gene expression. This intricate system of post-translational modifications is essential for various cellular processes, including DNA replication, repair, and recombination, in addition to its primary role in regulating gene activity.
