GMPAMPcGAMP

GMPAMPcGAMP is a cyclic dinucleotide (CDN) that plays a significant role in innate immunity. It is synthesized by cGMP-AMP synthase (cGAS) in response to the detection of cytosolic DNA. The presence of foreign DNA, such as from viruses or bacteria, triggers cGAS to produce GMPAMPcGAMP. This molecule then binds to and activates the stimulator of interferon genes (STING) protein.

Upon activation by GMPAMPcGAMP, STING translocates from the endoplasmic reticulum to the Golgi apparatus. This translocation initiates a signaling cascade that ultimately leads to the production of type I interferons (IFNs) and other pro-inflammatory cytokines. Type I interferons are crucial for establishing an antiviral state in neighboring cells, thereby limiting pathogen replication and spread. The GMPAMPcGAMP-STING pathway is a key component of the innate immune system, providing a rapid defense against intracellular pathogens. Dysregulation of this pathway has been implicated in various autoimmune and inflammatory diseases.
