Sphingomyelin-containing refers to any lipid or membrane that includes sphingomyelin as a constituent. Sphingomyelin is a major sphingolipid in animal cell membranes, characterized by a ceramide backbone linked to a phosphocholine headgroup. It is a structural lipid that contributes to membrane organization and the physical properties of the lipid bilayer.

Sphingomyelin is particularly abundant in the outer leaflet of the plasma membrane and is a prominent component of myelin sheaths around nerve fibers, where its presence helps create compact, stable membranes. It participates in the formation of lipid rafts—microdomains rich in cholesterol and sphingolipids—that organize signaling molecules and membrane trafficking processes. Through its metabolism, sphingomyelin also participates in cellular signaling pathways, notably via the release of ceramide when sphingomyelin is hydrolyzed.

Biosynthesis of sphingomyelin occurs mainly in the Golgi apparatus, where sphingomyelin synthase transfers a phosphocholine group to ceramide. In cells, sphingomyelin is degraded by sphingomyelinases to produce ceramide and phosphocholine; ceramide itself can act as a signaling lipid influencing processes such as apoptosis and differentiation.

Clinically, alterations in sphingomyelin levels or metabolism are linked to disease. Deficiency of acid sphingomyelinase causes Niemann-Pick disease types A and B, characterized by sphingomyelin accumulation in organs. Sphingomyelin-containing lipids are also studied in lipidomics and model membranes to understand membrane structure, trafficking, and signaling in health and disease.
