Kostimulációval refers to the process of providing a secondary signal in addition to the primary signal to achieve a biological response. In immunology, this concept is fundamental to the activation of T lymphocytes. T cells require two signals for full activation. The first signal, Signal 1, is initiated when the T cell receptor (TCR) recognizes an antigen presented by an antigen-presenting cell (APC) in the context of MHC molecules. However, this signal alone is insufficient to elicit a robust T cell response and can even lead to anergy, a state of unresponsiveness.

Signal 2, the kostimulatory signal, is provided by the APC through the expression of specific costimulatory molecules on its surface. These molecules bind to their corresponding receptors on the T cell. A classic and well-studied costimulatory pathway involves the interaction between CD28 on the T cell and B7 molecules (CD80 and CD86) on the APC. When both Signal 1 and Signal 2 are received, T cell proliferation, cytokine production, and differentiation into effector cells are effectively promoted.

Kostimuláció is crucial for ensuring that T cell activation is tightly regulated and occurs only in response to genuine foreign antigens, thereby preventing autoimmune reactions. Conversely, the absence or blockade of kostimulatory signals can lead to immune tolerance. Beyond T cell activation, the principle of kostimuláció is also observed in other biological contexts, where a combination of signals is needed for optimal cellular function. Understanding kostimulatory pathways has significant implications for the development of immunotherapies, such as cancer vaccines and treatments for autoimmune diseases.
