Karboksülaatioonist, also known as carboxylation, is a biochemical process where a carbon dioxide (CO₂) molecule is incorporated into an organic compound. This reaction is fundamental in various metabolic pathways, particularly in photosynthesis and the synthesis of essential biomolecules.

In photosynthesis, carboxylation occurs during the Calvin cycle, where the enzyme ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) catalyzes the fixation of CO₂ into a five-carbon sugar called ribulose-1,5-bisphosphate (RuBP). This reaction produces two molecules of 3-phosphoglycerate (3-PGA), which are later converted into glucose and other carbohydrates. Carboxylation is the first major step in carbon fixation, enabling plants to convert inorganic CO₂ into organic compounds necessary for growth and energy storage.

Beyond photosynthesis, carboxylation plays a role in the synthesis of amino acids, fatty acids, and other metabolites. For example, acetyl-CoA carboxylase catalyzes the carboxylation of acetyl-CoA to malonyl-CoA, a precursor in fatty acid biosynthesis. This process is critical for lipid metabolism and energy homeostasis.

Carboxylation reactions are also involved in the regulation of metabolic pathways. In some cases, enzymes like pyruvate carboxylase add CO₂ to pyruvate, forming oxaloacetate, which can enter the citric acid cycle or be converted into glucose via gluconeogenesis. This highlights the versatility of carboxylation in maintaining cellular energy balance and biosynthesis.

The efficiency and specificity of carboxylation depend on the enzyme involved, environmental conditions, and substrate availability. For instance, Rubisco’s dual function as a carboxylase and oxygenase influences photosynthetic efficiency, particularly under varying CO₂ and oxygen concentrations. Understanding carboxylation mechanisms helps in improving crop productivity, optimizing industrial bioprocesses, and developing strategies for carbon capture technologies.
