Gram-negative bacteria possess a thin peptidoglycan layer that is surrounded by an outer membrane containing lipopolysaccharides (LPS). This outer membrane acts as a barrier, preventing the crystal violet-iodine complex from reaching the peptidoglycan layer and being retained during the decolorization step.
